چکیده

The emergence of hybrid DNA–protein nanostructures in recent years has expanded the application DNA nanotechnology. Previous studies reported integration proteins into by sequence-dependent interactions or chemical modifications DNA, which limit design flexibility nanostructures. Here we report construction sequence-independent geometry-based using self-assembly single-stranded (ss) and histone proteins. We demonstrate that nucleosome-like ssDNA-histone complexes are integrated various shapes origami at specific predefined locations. further show possibility these as building blocks for designing more complex Our finding would facilitate development predesigned flexible straightforward higher-order structures.

Higher-order organization by mesoscale self-assembly and transformation of hybrid nanostructures.

The organization of nanostructures across extended length scales is a key challenge in the design of integrated materials with advanced functions. Current approaches tend to be based on physical methods, such as patterning, rather than the spontaneous chemical assembly and transformation of building blocks across multiple length scales. It should be possible to develop a chemistry of organized ...

DNA–melamine hybrid molecules: from self-assembly to nanostructures

Single-stranded DNA-melamine hybrid molecular building blocks were synthesized using a phosphoramidation cross-coupling reaction with a zero linker approach. The self-assembly of the DNA-organic hybrid molecules was achieved by DNA hybridization. Following self-assembly, two distinct types of nanostructures in the form of linear chains and network arrays were observed. The morphology of the sel...

DNA origami: synthesis and self-assembly.

DNA origami is an emerging technology for designing defined two- and three-dimensional (2D and 3D) DNA nanostructures. Here, we report an introductory practical guide with step-by-step experimental details for the design and synthesis of origami structures, and their size expansion in 1D and 2D space by means of self-assembly.

Lipid-bilayer-assisted two-dimensional self-assembly of DNA origami nanostructures

Self-assembly is a ubiquitous approach to the design and fabrication of novel supermolecular architectures. Here we report a strategy termed 'lipid-bilayer-assisted self-assembly' that is used to assemble DNA origami nanostructures into two-dimensional lattices. DNA origami structures are electrostatically adsorbed onto a mica-supported zwitterionic lipid bilayer in the presence of divalent cat...

DNA Self-Assembly For Constructing 3D Boxes

We propose a mathematical model of DNA self-assembly using 2D tiles to form 3D nanostructures. This is the first work to combine studies in self-assembly and nanotechnology in 3D, just as Rothemund and Winfree did in the 2D case. Our model is a more precise superset of their Tile Assembly Model that facilitates building scalable 3D molecules. Under our model, we present algorithms to build a ho...
